Summary
In the thymus of adult mice the number of cells in mitosis decreases markedly from the cortical to the medullary zone. After colchicine blockade the maximum is found in the sub-capsular cortical zone, where 20% of the cells show arrested mitoses. Most of the cells in this layer are poorly differentiated strongly basophil stem cells and lymphoblasts. After a single administration of 3H-thymidine 60% of these cells show the label in the nucleus. Because of cellular migration the majority of these labeled nuclei is seen at the cortical-medullary borderline within 24 hours. These cells stem from the sub-capsular cortical zone, which is the germinative layer of the cortex of the thymus. Only small numbers of 3H-labeled cells are seen in the centre of the medulla. They are found in great numbers on and in the perilymphatic sheaths of the blood vessels in the cortical-medullary border region. 48 hours p.i. a distinct decrease in the number of labeled cells as well as in the amount of activity is observed. This observation finds its explanation in the emigration of continuously newly formed cells from the thymus.
Zusammenfassung
Im Thymus der erwachsenen Maus hat die Mitosehäufigkeit ein starkes Gefälle von der Außenzone der Thymusrinde zum Mark. Nach Colchizinblockade liegt das Maximum in der subkapsulären Randzone, wo 20% der Zellen als blockierte Mitosen vorliegen. Die Zellen dieser Schicht sind überwiegend wenig differenzierte stark basophile Stammzellen und Lymphoblasten. Nach einmaliger Gabe von 3H-Thymidin findet sich eine Markierung der Kerne von 60% dieser Zellen. Innerhalb von 24 Std verlagert sich das Maximum der Kernmarkierung durch Zellwanderung an die Rinden-Markgrenze. Ursprungsort dieses Zellstromes ist die subkapsuläre Randzone, die Keimschicht der Thymusrinde. 3H-markierte Zellen treten nur in geringem Maße in das innere Mark über. Sie sind gehäuft an und in den Perilymphscheiden der Gefäße der Rinden-Markgrenze nachzuweisen. 48 Std p.i. tritt eine erhebliche Abnahme beladener Zellen und ihres Beladungsgrades ein. Sie kann nur mit einer Ausschleusung ständig neu gebildeter Zellen aus dem Thymus erklärt werden.
