Zusammenfassung
Unsere kombinierten Untersuchungen des experimentellen Osteoangiolathyrismus junger Schweine haben eine veränderte Zusammensetzung der Grundsubstanz bestätigen können. Der Gehalt an sauren Mucopolysacchariden ist im Knorpel und in der Aorta verringert.
Diese generalisierte lathyrogene Alteration des Bindegewebes ist Ausdruck einer Hemmung der Biosynthese der sauren Mucopolysaccharide in den ortsständigen Bindegewebszellen.
Die Lathyrusvergiftung ist theoretisch erklärbar als Enzymblockade. Am Beispiel des Transaminierungsvorganges mit Hilfe des Coenzyms Pyridoxal erläutern wir diese Vorstellung für Aminonitrile, Hydrazinabkömmlinge und CH-acide Verbindungen. Unter der Annahme eines Mangels an sauren MPS durch die Hemmung der Biosynthese werden Befunde aus der Literatur diskutiert. Zusammenfassend kann man folgendes Schema für die BAPN-Wirkung am Bindegewebe und an der Aorta aufstellen:

Stop der Biosynthese der sauren MPS nach Applikation von Lathyrusdrogen abhängig von der Dosis im Sinne einer Enzymblockade.

2.
Verarmung der Interzellularsubstanz und auch der Fibrillen an sauren MPS.

3.
Störung der Fibrillogenese in der Interzellularsubstanz.
-
4.
Herabsetzung der „äußeren und inneren Stabilität“ der kollagenen und elastischen Fasern und Fibrillen; Depolymerisation.

5.
Hydropische Quellung des Bindegewebes und der Aorta durch eine veränderte Elektrolytzusammensetzung.

6.
Indirekte Schädigung des Erhaltungsstoffwechsels der Bindegewebsfibrillen und der glatten Muskelzellen der Aortenwand durch Verlängerung des Diffusionsweges und Fehlen der sauren MPS als Ionentransporter in der Interzellularsubstanz.

7.
Störung der Mineralisation in der Wachstumszone des Rippenknorpels; Entstehung eines rachitisartigen Zustandsbildes.

8.
Deutung der Mineralisationsstörung als Folge der Aufhebung der Induktion einer Kristallbildung durch reifes Kollagen, da eine Ausreifungsstörung und Depolymerisation der kollagenen Fasern vorliegt.
