Summary
The taste buds on the barbels of Amiurus nebulosus were investigated morphologically and histochemically.
Light microscopic investigations: The pear-shaped taste buds being situated in the epidermis of the barbels contain several cell types:

a)
Marginal cells forming the boundary to normal epithelial cells of the epidermis.

b)
Sensory cells in the center of the organ.

c)
Basal cells located at the basis of the organ.
-
d)
A nerve plexus is situated between the sensory cells and the basal cells.
As demonstrated by fluorescence microscopy only the basal cells contain an aminergic transmitter, the serotonin (5-hydroxytryptamine). An activity of the enzyme monoaminooxidase also exists only in the basal cells. On the other hand, the activity of acetylcholine esterase is restricted to the nerve plexus. The sensory cells cannot be identified by any of these methods.
Electron microscopic investigations: The sensory cells can be divided into 1) light sensory cells (with a clear cytoplasma, well developed membranes of the smooth endoplasmatic reticulum, a compact system of microtubules and apically with one great microvillus) and 2) dark sensory cells (with a dense cytoplasma, dense secretory granules, only a few microtubules and with a lot of filaments). While the light sensory cells are ending at the level of the nervous plexus, the processes of the dark sensory cells penetrate the plexus and reach the basal cells as well as the basal membrane of the papilla of the corium. The basal cells have a clear cytoplasm with numerous mitochondria and vesicles of the smooth endoplasmatic reticulum.
Synapses are situated between sensory cells (presynaptic side; p.s.) and nerve fibers, between sensory cells (p.s.) and basal cells and between basal cells (p.s.) and the fibers of the nervous plexus. After fixation with KMnO4 in the sensory cells only clear synaptic vesicles (ca. 300 Å in diameter) are found. However, the basal cells contain clear and electron dense vesicles of the same size. The dense core vesicles probably contain serotonin (5-hydroxytryptamine; argentaffine reaction). An activity of acetylcholine esterase is found on the nervous plexus and on the basal part of the sensory cells. The reaction product lies between the nerve fibers and between nerve fibers and sensory cells.
The possibilities of impulse transmission within the taste bud are discussed. It may be possible, that a) an impulse is directly reaching the CNS via an afferent, cholinergic nerve, and that b) an impulse is first transmitted to a basal cell in a cholinergic manner, where the impulse is intensified or is coordinated with other impulses and that it is then transmitted to the CNS in an afferent, but aminergic nerve, too.

Die auf den Barteln von Amiurus nebulosus liegenden Geschmacksknospen wurden morphologisch und histochemisch untersucht.
Lichtmikroskopie: Die birnförmigen Geschmacksorgane in der Bartel-Epidermis werden von mehreren Zelltypen aufgebaut; es sind dies:

a)
Am Rand liegende Übergangszellen, die die Grenze zum Körperepithel bilden.
-
b)
Im Innern des Organs liegende Sinneszellen.
-
c)
An der Basis des Organs gelegene Basalzellen. — Zwischen den Sinneszellen und den Basalzellen liegt d) ein Nervenfaserplexus.
Der fluoreszenzmikroskopische Nachweis biogener Amine ergab, daß lediglich in den Basalzellen ein aminerger Transmitter, das Serotonin (5-Hydroxytryptamin), vorkommt. Auch sind die Basalzellen die einzigen Orte, die eine Monoaminoxidase-Aktivität zeigen. Die Aktivität der Acetylcholinesterase ist dagegen auf den Bereich des Nervenfaserplexus beschränkt. Die Sinneszellen lassen sich mit keiner der angewandten Methoden darstellen.
Elektronenmikroskopie: Die Sinneszellen lassen sich in 1) helle Sinneszellen (helles Cytoplasma, stark entwickelte Membransysteme des glatten Endoplasmatischen Reticulum, dichtes Mikrotubuli-System, kräftiger apikaler Mikrovillus) und 2) dunkle Sinneszellen (dichtes Cytoplasma, elektronendichte Sekretgranula, weniger Mikrotubuli, mehr Filamente) einteilen. Während die hellen Sinneszellen im Bereich des Nervenfaserplexus enden, durchdringen die dunklen Sinneszellen mit ihren Fortsätzen den Plexus und erreichen die Basalzellen bzw. die Basalmembran der Coriumpapille. Die Basalzellen erscheinen hell; ihr Zytoplasma ist reich an Mitochondrien und Vesikeln des glatten endoplasmatischen Retikulum.
Synapsen liegen zwischen Sinneszellen (präsynaptische Seite; p.S.) und Nervenfasern, zwischen Sinneszellen (p.S.) und Basalzellen und zwischen Basalzellen (p.S.) und Nervenfasern des Plexus. Nach KMnO4-Fixierung sind in den Sinneszellen lediglich helle synaptische Vesikel (ca. 300 Å Durchmesser) nachweisbar; in den Basalzellen dagegen helle und elektronendichte Vesikel gleicher Größe. Die dichten Vesikel beinhalten sehr wahrscheinlich Serotonin (Argentaffinitätsreaktion). Die Aktivität der Acetylcholinesterase beschränkt sich weitgehend auf den Nervenfaserplexus und die Basis der Sinneszellen, wobei das Reaktionsprodukt zwischen den Nervenfasern bzw. zwischen Nervenfasern und Sinneszellen liegt.
Aufgrund der morphologischen Befunde werden die Möglichkeiten der Erregungsübermittlung im Bereich der Geschmacksknospe diskutiert. Es erscheint denkbar, daß

a)
eine Erregung von den Sinneszellen über afferente Bahnen direkt zum ZNS geleitet wird (cholinerg), und daß
-
b)
eine Erregung zunächst den Basalzellen übermittelt wird (cholinerg), die sie ver stärken oder mit anderen Erregungen koordinieren, bevor sie über afferente Bahnen an das ZNS weitergeleitet wird (aminerg).
