Summary
The ultrastructure of the zona radiata and adjacent tissues of growing oocytes of several salmonids and of Fundulus heteroclitus has been investigated by light and electron microscopy. In all species investigated so far the zona radiata consists of two layers, the electron dense zona radiata externa, and the slightly less dense zona radiata interna. Both layers are traversed by numerous pore canals. The osmiophilic “externa” of more mature oocytes of the trout encircles the outer openings of the canals, thus forming well defined “pore openings”. Each pore canal contains one microvillus of the oocyte. As growth continues, processes of the follicular cells penetrate the pore canals, where they are in close contact with the microvilli. Shortly before ovulation both the microvilli and the follicular processes are withdrawn from the canals. Simultaneously, the “externa” forms plugs within the outer openings of the canals, thus closing them completely. The dots often noted on the surface of the “externa” are caused by these plugs. The remaining canals are responsible for the striated appearance of the “interna” of salmonids as seen with the light microscope. In Fundulus the “interna” slggests a structural framework. Already before ovulation the framework of the “interna” turns into the solid capsule of mature eggs. This process commences at the inside of the “interna”. The follicular processes leave the canals, followed by the withdrawal of the microvilli. The pore canals are closed over their entire length, possibly by addition of material to their inner walls. The follicular cells of all species investigated are separated by intercellular spaces of different size and shape. Osmiophilic strands arising from the “externa” are present within the intercellular spaces of the Fundulus follicle. These strands are composed of a highly organized material as revealed by the examination of extremely thin sections. Just before ovulation the intercellular spaces disappear, apparently as a result of the withdrawal of the follicular processes. Adjacent cells again make close contact with one another. During the ovulation of the eggs of Salvelinus and of Salmo the follicular epithelium together with the subfollieular layer comes off, thus shedding the mature egg. Not until after ovulation a layer of jellylike material is added. The eggs of Fundulus are not covered with a jelly layer. After the induced degeneration of oocytes, electron dense bodies and lamellae are formed within the ooplasm and the cytoplasm of the follicular cells. These are believed to be lysosomes. The microvilli as well as the follicular processes are withdrawn prematurely.
Zusammenfassung
Die Feinstruktur der Zona radiata und angrenzender Gewebe wurde an wachsenden Oozyten und Eiern verschiedener Salmoniden und Fundulus heteroclitus licht- und elektronenmikroskopisch untersucht. Bei allen bisher untersuchten Arten besteht die Zona radiata aus zwei Schichten, der elektronendichten Zona radiata externa und der kontrastärmeren Zona radiata interna. Beide Schichten werden von Kanälen perforiert. Die osmiophile „Externa“ der weiter entwickelten Oozyten der Salmoniden umschließt ringförmig die äußeren Öffnungen der Kanäle. Jeder Porenkanal enthält einen Mikrovillus der Oozyte. Etwas später dringen Fortsätze der Follikelzellen in die Porenkanäle ein, wo sie in engem Kontakt mit den Mikrovilli liegen. Kurz vor der Ovulation weichen die Mikrovilli und die follikularen Fortsätze zurück, die Wandungen der Porenöffnungen verwachsen zu den typischen „Verschlußpfropfen“ und verschließen die Kanäle. Die Porenkanäle bleiben erhalten und verursachen bei Salmoniden die bekannte Streifung der Zona radiata interna lichtmikroskopischer Untersuchungen. Die Pfropfen sind für das Punktmuster auf der „Externa“ verantwortlich. Die „Interna“ der Funduluseier besteht aus einem Fachwerk. Schon vor der Ovulation werden die Zwischenräume des Fachwerkes ausgefüllt. Dieser Vorgang beginnt innen und schreitet unter gleichzeitigem Zurückweichen der Fortsätze der Follikelzellen aus den Porenkanälen nach außen fort. Zum Schluß verlassen auch die Mikrovilli die Kanäle, die dann in ihrer ganzen Länge verschlossen werden. Das Follikelepithel aller untersuchten Arten ist während der Oogenese von Interzellularspalten wechselnder Form und Größe separiert. Bei Fundulus liegen in diesen Interzellularspalten osmiophile Stränge, die einen lamellären Feinbau aufweisen. Vor der Ovulation werden die Interzellularspalten zunehmend zurückgebildet. Während der Ovulation der Eier von Salmo und Salvelinus löst sich mit dem Follikelepithel die subfollikulare Schicht von der Zona radiata externa. Nach der Ovulation erhalten die Eier dann eine Gallerthülle. Funduluseier haben keine Gallerthülle. Nach induzierter Degeneration der Oozyten werden elektronendichte Körperchen und lamelläre Cytosomen im Ooplasma der Follikelzellen gebildet. Gleichzeitig werden die Mikrovilli sowie die Fortsätze der Follikelzellen vorzeitig aus den Porenkanälen zurückgezogen.
