On the mechanism of mitotic recombination in Aspergillus nidulans

II. Simultaneous recombination within two closely linked cistrons

Summary

Simultaneous double recombination between the ad9 alleles and the paba1 alleles was studied. Two types of polarity can be observed among mitotic recombinants selected for adenine- and/or pABA-independence: firstly, among selected pABA-independent recombinants there are twice as many double ad + paba + recombinants, as among selected for adenine-independence recombinants. Secondly, in both cistrons studied, the distal ad and paba mutants converted to their respective wild-type alleles with higher frequency than did the proximal ad and paba mutants.

When multiple recombinants are selected, in the presence of spanning markers, the distinction between reciprocal and non-reciprocal events becomes blurred: the most reasonable assumption seems to be that a great majority or even all cross-overs are non-reciprocal over very short segments of genetic material.

Comparison of spanning marker distribution among selected mitotic and meiotic recombinants between the same pairs of paba mutants suggests that the marker distribution can be influenced by elements of chromosome structure other than nucleotide sequences in DNA.
