A new locus, stc, which affects the phenotype of tolC mutants of Escherichia coli K-12

Summary

Mutants at the tolC locus in Escherichia coli K-12 do not express the OmpF protein. We sought revertants which do produce this protein in a tolC background. We obtained only one type, termed stc (suppressor of tolC). stc mutations had no effect on the other components of the tolC phenotype and mapped at 47.5 min on the genetic map. Some interaction between stc and ompC was detected, but this may be indirect and due to a separate regulation of total porin levels. A second class of suppressor mutation studied was the ompR independent mutation, ompFp9, described by Sato and Yura (1981). The stc and ompFp9 mutations can have very similar effects on the phenotype, but whereas ompFp9 is thought to be a constitutive mutation which is no longer dependent on ompR positive control, stc either acts at a later stage, in transcription, or posttranscriptionally.
