Abstract
Isolated salivary gland chromosomes of Chironomus tentans incubated in a simple salt or sucrose solution are able to synthesize not only RNA but also protein in their puffs. The migration of radioactively labelled material from isolated nucleoli to isolated chromosomes suggests that not all protein in the puffs is synthesized by the puffs themselves but that part of the puff protein stems from the nucleoli. It is concluded that besides bound RNA polymerase a puff contains ribosomes attached to the growing mRNA molecules.

Die Fähigkeit isolierter Speicheldrüsen-Chromosomen von Chironomus tentans zu autonomer RNS- und Protein-Synthese wurde autoradiographisch untersucht.

3H-markiertes Cytidin-triphosphat (3H-CTP) wird vorwiegend in die RNS der puffs, im speziellen der Balbianiringe, eingebaut. Zu dieser RNS-Synthese in den puffs bedarf es nichts anderes als der 4 Ribonukleosid-triphosphate, gelöst in einem einfachen Salz-bzw. Zuckermedium. Puffs scheinen demnach festgebundene RNS-Polymerase zu enthalten (vgl. „Aggregat-Enzym“, S. B. Weiss, 1960).

Andauung mit Trypsin verändert das RNS-Synthese-Muster isolierter Chromosomen nicht. Daraus wird geschlossen, daß Chromosomen-Regionen außerhalb der puffs weniger oder keine RNS-Polymerase besitzen.

14C-markierte Aminosäuren werden ebenfalls vor allem in den gepufften Chromosomen-Regionen inkorporiert. Es wird deshalb angenommen, daß puffs, außer zur autonomen RNS-, auch zur autonomen Protein-Synthese fähig sind, woraus geschlossen wird, daß puffs Ribosomen enthalten.

Isolierte Chromosomen wurden gemeinsam mit isolierten radioaktiv-markierten Zellbestandteilen inkubiert. Aus dem Kernsaft und den Nukleolen wandert markiertes Protein in die Chromosomen und in die puffs. Es wird vermutet, daß der aus den Nukleolen stammende Teil des Proteins in den puffs dem Struktur-Protein von Ribosomen entspricht.

Auf Grund der Ergebnisse der vorliegenden Arbeit wird ein Funktionsmodell eines puff aufgestellt.
