An Feulgen-gefärbten Speicheldrüsen-Chromosomen des Unterart-Bastards von Chironomus thummi wurden 30 verschiedene Querscheiben auf subspezifische Differenzen des DNS-Gehalts cytophotometrisch untersucht. Mit der gleichen Technik wurde die Höhe von DNS-Differenzen an den heterochromatischen Kinetochorscheiben der Speicheldrüsen-Chromosomen von Chironomus plumosus bestimmt.

Bei subspezifischer Differenz des DNS-Gehalts enthalten die Querscheiben von Ch. thummi thummi das 2-, 4-, 8- oder 16fache an DNS wie ihre Homologen bei Ch. thummi piger. Die innerhalb der Subspecies Ch. thummi thummi und der Species Ch. plumosus vorkommenden Strukturtypen bestimmter Querscheiben folgen in der Differenzierung ihres DNS-Gehaltes dem gleichen Prinzip.

Der relative Gesamt-DNS-Gehalt der Speicheldrüsenkerne von Ch. th. thummi ist um 27% höher als derjenige von Ch. th. piger. Die gleiche Differenz des DNS-Gehalts ist auch in den Spermatocyten I cytophotometrisch nachweisbar.

Nach den Untersuchungen über die phylogenetischen Beziehungen innerhalb der Gattung Chironomus (Keyl 1962a) hat sich der Karyotyp von Ch. th. thummi aus demjenigen von Ch. th. piger entwickelt. Demnach beruht die Differenzierung der Chromosomen beider Unterarten auf einer lokalen Zunahme des DNS-Gehalts.

Auf Grund der cytophotometrischen Resultate und der Befunde über das DNS-Synthese-Muster in den Speicheldrüsen-Chromosomen von Ch. thummi (Keyl und Pelling 1963) wird eine Hypothese entwickelt, nach der die lokale Zunahme des DNS-Gehalts in den Chromosomen auf einer schrittweisen Erweiterung von DNS-Replikationseinheiten im 1∶2-Verhältnis beruht. Es wird angenommen, daß bei jedem Verdoppelungsschritt das in der Replikationseinheit vorhandene DNS-Stück um eines von gleichem genetischen Inhalt verlängert wird.

Aus dem während der Evolution sich wiederholenden Duplikationsvorgang wird auf Gestalt und Anordnung der Replikationseinheiten geschlossen und ein Ablaufschema des Duplikationsmechanismus erörtert.

The DNA content of 30 selected pairs of homologous bands in the unpaired regions of the salivary gland chromosomes of the subspecies hybrid Chironomus th. thummi × Ch. th. piger has been measured with a Zeiss ultramicrospectrophotometer. With the same technique certain heterochromatic bands in the vicinity of the kinetochore of Ch. plumosus have been investigated.

Some bands of Ch. th. thummi contain 2, 4, 8 or 16 times the amount of DNA as compared to the homologous bands in Ch. th. piger. Quantitative steps of the same type have been found within different populations of Ch. th. thummi and in Ch. plumosus.

The total amount of DNA in salivary gland nuclei of Ch. th. thummi is 27% higher than in Ch. th. piger. The same relative difference has been measured in first spermatocytes.

From phylogenetic relations within the genus Chironomus (Keyl, 1962a) it is to be concluded that Ch. th. thummi is derived from Ch. th. piger. The difference in their chromosomes thus is caused by localized increase of DNA in Ch. th. thummi.

From these cytophotometric results and the way of replication in bands of different thickness (Keyl and Pelling, 1963) the hypothesis is formulated that the discovered type of DNA increase results from a stepwise elongation of replication units. In each step a whole unit is added whereby a new unit of double length originates. The new unit again can be doubled resulting in a unit of four times the DNA amount of the original unit.

A scheme is proposed (Fig. 11) for a possible mechanism of the doubling of the units by errors in replication.
