Summary
44 male guinea pigs were injected with 6 mg/kg cortisone, alternatively 18 mg/kg hydrocortisone, daily over a period of 28 days. A reduction of ACTH secretion was ascertainable by numerical decrease of the R-cells in the anterior lobe of the hypophysis and by the “regressive transformation” of the adrenal cortex, as observable in histological preparations.
The diminished secretory activity of the adenohypophysis is reflected by morphological reactions in the nucleus infundibularis, nucleus suprachiasmaticus, nucleus paraventricularis thalami and in the hippocampus. The alterations, i.e. the irreversible pyknosis of the nerve cell nuclei and the enlargement of the remainder in the same area, were independent of the type kind of glucocorticoid administered. After intensive stress, comparable lesions appeared in the three aforementioned diencephalic nuclear areae. By means of the sodium-hypochlorite-Schiff reaction, the pyknotic nuclei and the intact ganglionic cells of all four of the nerve cell regions referred to could be selectively demonstrated as is the case with nerve cells in these regions of normal animals. By employing the coupled tetrazonium reaction, only the shrunken cell nuclei were stained.
The results confirm the significance of these regions in the control of the adrenocorticotropic partial function of the adenohypophysis.
Zusammenfassung
44 männliche Meerschweinchen erhielten 28 Tage lang 6 mg/kg Cortison bzw. 18 mg/kg Hydrocortison. Die dadurch bedingte Herabsetzung der ACTH-Sekretion konnte durch die verminderte Zahl der R-Zellen im Hypophysenvorderlappen und die „regressive Transformation“ der Nebennierenrinde histologisch nachgewiesen werden.
Die reduzierte sekretorische Aktivität der Adenohypophyse zog morphologisch faßbare Reaktionen im Nucleus infundibularis, Nucleus suprachiasmaticus, Nucleus paraventricularis thalami und im Hippocampus nach sich. Die Veränderungen in Form von irreversiblen Kernpyknosen und reversiblen Vergrößerungen der restlichen Nervenzellkerne innerhalb eines Kerngebietes waren von der Art des verabfolgten Glucocorticoids unabhängig. Nach intensivem Stress traten vergleichbare Schädigungen in den drei genannten Kerngebieten des Zwischenhirns auf. Durch die Natriumhypochlorit-Schiff-Reaktion kamen die pyknotischen Zellkerne und die ungeschädigten Ganglienzellen der vier Nervenzellregionen selektiv zur Darstellung; auch bei Normaltieren sind die Nervenzellen dieser Gebiete mit der Natriumhypochlorit-Sehiff-Reaktion färbbar. Mit der Tetrazonium-Kupplungsreaktion färbten sich nur die geschrumpften Zellkerne.
Die Ergebnisse bestätigen die Bedeutung der aufgeführten Areale für die Kontrolle der adrenocorticotropen Partialfunktion des Hypophysenvorderlappens.
