Summary
The insulin release of in vitro grown endocrine rat and pig pancreatic cells was not influenced by colchicine. The cells (with or without treatment) contained the same amount of insulin.
After changing the nutritive medium the insulin content of the cells was decreased by the corresponding quantity of insulin released into the respective medium. Addition of caffeine increased the insulin release in experimental as well as in control cultures in the same manner, whereas secretin had no influence.
Morphological analyses (light microscope, scanning microscope, slow motion film) revealed structural alterations in cytoplasm and cell surface. This we consider as an expression of the reaction between colchicine and microtubular protein.
Zusammenfassung
Der Insulinrelease in vitro gezüchteter Ratten- („special cells“) und Schweinepankreaszellen, die aufgrund der Aufarbeitungsmethode an endokrinen Zellen angereichert sind, bleibt durch Colchicin unbeeinflußt. Die Zellen selber (behandelt oder unbehandelt) enthielten die gleichen Mengen Insulin.
Nach Wechsel der Nährlösung verringerte sich ihr Insulingehalt um die in die Nährlösung abgegebene Insulinmenge. Koffeinzusatz vermehrte bei Versuchs- und Kontrollkulturen den Insulinrelease gleichartig, während Sekretin in beiden Fällen keinen Einfluß hatte.
Morphologische Analysen (Lichtmikroskop, Zeitrafferfilm, Stereoscan®) ergaben Strukturveränderungen im Cytoplasma und an der Zelloberfläche, die als Ausdruck der Reaktion zwischen Colchicin und mikrotubulärem Protein gedeutet werden.
