Zusammenfassung

Mit Dichlorpseudocyanin (8,6 mg/100 ml Wasser) lassen sich bei Formol/ Sublimat- oder Helly-fixierten, mit Perameisensäure oxydierten Schnitten im Hypophysenvorderlappen der Ratte die thyreotropinbildenden Zellen durch eine kräftige metachromatische Reaktion elektiv darstellen. Diese Reaktion ist bei monochromatischem Licht (578 nm) und fluoreszenzmikroskopisch besonders gut erkennbar und entspricht dem Ausfall der Perameisensäure-Alcianblau-Färbung.

Die Reaktion ist auch an den Hypophysen vom Menschen und von anderen Spezies mit Erfolg durchführbar, wobei die funktionelle Deutung der dargestellten Zellen hier noch offen bleibt.

Die Metachromasie wird auf die Anwesenheit von Cystin zurückgeführt, dessen Disulfidgruppen zu Sulfogruppen oxydiert werden, die eine Polymerisation der Dichlorpseudoisocyaninmoleküle bewirken.

The thyrotropin-secreting cells of the rat pituitary (fixation: formalinsublimate or Helly) can be stained selectively by means of a strong metachromatic reaction with dichlorpseudoisocyanine (8,6 mg/100 ml water) after oxidation in performic acid. The reaction can be detected most distinctly in monochromatic light (578 nm) or by fluorescence microscopy, and is in accordance with the result of the performic acid-Alcian blue staining.

The staining is successful also in the anterior pituitary of man and several other species; the functional interpretation of these results remains open.

The metachromasia is attributed to the presence of cystine, the disulphide groups of which are oxidized to sulpho groups and thus effect a polymerisation of the dichlorpseudoisocyanine molecules.
