Summary
Smeras of liver parenchyme cells of the white mouse were subjected to the Feulgen reaction (PNB) using normal Schiff's reagent. Cytophotometric measurements yielded a standard karyogram (showing 2n, 4n, 8n, and 16n nuclei) with which the other ones were compared. —From a great number of basic (cationic) dyes of different chemical nature some few proved suitable to prepare the so-called Schiff-type reagents, viz. Safranin 0, Neutral red, Coriphosphine 0, Toluidine blue 0, and Cresyl violet. The absorption maxima of the colours developed in the nuclei on the one hand and of those of the respective aqueous solutions of the pure dyes on the other differ clearly in the case of Neutral red, Toluidine blue and Cresyl violet, as it is the case in the Feulgen colour and in the aqueous solution of basic Fuchsin. This indicates that the colour of the nuclei (as in the FNR!) does not originate from a simple restoration of the dye decolorized. (In Coriphosphine and Safranin the micro- and macrophotometric curves were nearly identical.) However the observation of the colour development in the preparations indicates a staining mechanism different from that in the FNR. In control preparations (a. hydrolysis omitted, b. DNA quatitatively extracted) the Schifftype reaction is wanting. Thus, a specificity for DNA may be stated.—The liver cell nuclei stained by the five Schiff-type reagents yielded very similar karyograms as in the case of the FNR. Quantitative cytophotometry is therefore possible.—The question whether the Schiff-type reactions are possibly more intensive in colour than the FNR (whereby even lower DNA concentrations came to be measurable) must be answered in the negative (testing objects: slices of oocyte nuclei of fishes, newts and mouses).
Zusammenfassung
Ausstriche von Leberparenchymzellkernen der weißen Maus wurden der Feulgen-Reaktion (FNR) unterworfen (normales Schiffsches Reagenz). Cytophotometrische Messungen ergaben ein Standard-Karyogramm (mit 2n-, 4n-, 8n und 16n-Kernen), mit dem die anderen Karyogramme verglichen wurden. — Aus einer großen Zahl von basischen (kationischen) Farbstoffen verschiedener chemischer Natur erwiesen sich einige wenige als geeignet zur Herstellung der sog. „Schiff-Typ“-Reagenzien, nämlich Safranin 0, Neutralrot, Coriphosphin 0, Toluidinblau 0 und Kresylviolett. Die Absorptionsmaxima der in den Kernen entwickelten Farben einerseits sowie diejenigen der entsprechenden wäßrigen Lösungen der reinen Farbstoffe andererseits unterscheiden sich deutlich bei Neutralrot, Toluidinblau und Kresylviolett, wie es auch beim Feulgen-Farbstoff und der wäßrigen Fuchsin-Lösung der Fall ist. Dies zeigt an, daß die Farbreaktionen (wie auch bei der FNR) nicht durch eine einfache Restaurierung der entfärbten Ausgangsfarbstoffe zustande kommen. (Bei Coriphosphin und Safranin sind allerdings die mikround die makrophotometrischen Kurven nahezu identisch.) Jedoch läßt die Beobachtung der Farbentwicklung in den Präparaten auf einen anderen Färbemechanismus schließen als in der FNR. In Kontrollpräparaten a) ohne Hydrolyse, b) nach DNS-Extraktion bleibt die Schiff-Typ-Reaktion aus. Die Spezifität für DNS ist also gesichert. — Die mit den fünf Schiff-Typ-Reagenzien gefärbten Leberzellkerne ergeben sehr ähnliche Karyogramme wie im Falle der FNR. Die quantitative Auswertung ist also gewährleistet. — Die Frage, ob die Schiff-Typ-Reaktionen möglicherweise farbintensiver sind als die FNR (wodurch noch geringere DNS-Konzentrationen meßbar würden), muß verneint werden (Testobjekte: Schnitte von Oocytenkernen von Fischen, Molchen und Mäusen).
