Abstract

A modified method for examination of chromosomes of meiotic cells in mammalian testes is described. A cell suspension is prepared from the tubules of the whole testis by use of hyaluronidase and stirring for 20 to 30 minutes. Using standard procedures this suspension is worked up into air—dried preparations. The method gives a very high yield of good pictures of several meiotic stages, especially of first and second metaphases and diakinesis, but also of diplotene. The slides are evaluated using a Zeiss-photomicroscop combined with a Zeiss-drawingapparatus. This combination makes use of the advantage of drawing as well as photographing without change of the microscopic field.
