Summary
From renal cortex of rats a fraction of basement membranes could be enriched by means of non-continuous or layer gradient centrifugation with sucrose. The purity of this fraction and the fine structure of isolated basement membranes was examined by phase contrast and electron microscopy. They consist of a network of fine filaments being embedded by a homogeneous matrix. The collagen content of the examined fraction amounted to 28,8 per cent (dry weight). It is assumed the fine filaments being build up by collagen structures. Furthermore 39,6 per cent noncollagen protein and 16.9 per cent total lipides could be found. In contrast no mucopolysaccharides containing uronic acids could be detected.
Zusammenfassung
Aus Nierenrinden von Ratten konnte durch eine nichtkontinuierliche oder Schichtengradienten-Zentrifugation in Saccharose eine Basalmembranfraktion angereichert werden. Durch phasenkontrastund elektronenmikroskopische Betrachtung wurde die Reinheit dieser Fraktion geprüft und die Feinstruktur der isolierten Basalmembranen dargestellt. Sie bestehen aus einem Netzwerk feiner Filamente, die in einer homogenen Matrix eingelagert sind. Der Kollagengehalt in den untersuchten Fraktionen beträgt 28,8 % der Trockensubstanz. Die feinen Filamente werden deshalb für Kollagenstrukturen gehalten. Daneben konnten noch 39,6% nicht-kollagene Proteine und 16,9% Gesamtlipoide gefunden werden. Uronsäurehaltige Mukopolysaccharide konnten nicht nachgewiesen werden.
