The chemical determination of section thickness

Summary

In quantitative histochemistry it is frequently desirable to know the exact thickness of the sections. The nucleic acid content of the tissue has been used as the basis of a method for determining section thickness. Such determinations agree remarkably well with the setting on the cryostat microtome although considerable variations in thickness can occur in sections cut at different speeds.
