The efficacy of various mutagens and polyene antibiotics for the induction and isolation of sterol mutants of Neurospora crassa

Summary

Four mutagens and four polyene antibiotics were compared with respect to the induction and isolation of mutants carrying gene mutations affecting the biosynthesis of ergosterol. Almost 200 sterol mutants were isolated and classified into five groups on the basis of their growth characteristics, levels of antibiotic resistance and UV absorption spectra of sterol extracts. The most effective mutagen was the acridine “mustard”, ICR-170, and mutants were isolated most readily on media containing the polyene antibiotic, nystatin. Two genes affecting sterol biosynthesis and polyene resistance have been located on chromosome V.
