Repair of gamma-ray induced lesions in E. coli cells deficient in DNA polymerase I and having thermosensitive DNA polymerase III

Summary

A 2 hr-incubation of gamma-irradiated E. coli polA1 tsdnaE at 45° prior to plating at 30° results in the decrease of the fraction of viable bacteria. This effect is not observed in experiments with gamma-ray sensitive polA1 dnaE + and gamma-ray resistant polA1 + tsdnaE bacteria. The study of uptake of H3-thymine or H3-uridine into DNA or RNA of polA1 tsdnaE and polA1 dnaE + cells shows that the decrease in survival at 45° in the first strain after gamma-irradiation correlates with the DNA synthesis block rather than with the unspecific inhibition of RNA synthesis. DNA breakdown in gamma-irradiated polA1 tsdnaE cells was found to be higher at 45° as compared with 30°. The results may be interpreted as indicative of the participation of DNA-polymerase III in the repair of gamma-ray induced lesions possibly during replication.
