Some knowledge was obtained by electron microscopy about the kind of damage of Toxoplasma at low temperatures.
In rapid freezing of Toxoplasma probably water penetrates into the cell through the conoid by reason of pressure differences between the interior of the cell and its environment. By freezing of this water the cell then is ruptured.
Slow freezing causes damage of Toxoplasma by dehydration of the cell, whereas mechanical destructions of the cytoplasma are effected by recrystallization of the ice during thawing.
Addition of glycerol to the medium of suspension diminishes the dimensions of destructions and enables surviving of some Toxoplasma.
Zusammnfeassung
Auf Grund elektronenoptischer Beobachtungen an unterschiedlich eingefrorenen Toxoplasmen konnten einige Erkenntnisse über die Art der Schädigung dieser Erreger durch tiefe Temperaturen gewonnen werden.
Schnelles Einfrieren der Toxoplasmen führt zu erheblichen Strukturveränderungen. Dabei dringt wahrscheinlich infolge von Druckdifferenzen zwischen dem Inneren der Zelle und der Umgebung Wasser über das Conoid ein und führt durch anschließendes Gefrieren zu einer Ruptur der Zelle.
Beim langsamen Einfrieren kommt die Schädigung der Erreger wie bei anderen Zellen durch eine Dehydrierung zustande, während die mechanischen Zerstörungen des Plasmas durch Rekristallisation des Eises beim Auftauen erfolgen.
Ein Glyzerolzusatz zum Suspensionsmedium verhindert einen Teil der beschriebenen Schäden und ermöglicht damit einigen Toxoplasmen das Überleben.
