Zusammenfassung
Geschildert wird die Identifizierung, Darstellung und Charakterisierung eines präzipitierenden „funktionellen“ Fasciola hepatica-Antigens aus Totalhomogenatextrakten adulter Leberegel:
Seren experimentell infizierter Kaninchen präzipitierten in der Immunoelektrophorese 5–7 Totalhomogenatextrakt-Individuen. — Zwei dieser antigenen Komponenten verdienen besondere Beachtung wegen der Intensität des Präzipitationsvermögens und der im Vergleich mit den anderen „funktionellen“ Totalhomogenatextrakt-Antigenen frühesten Antikörperbildungspotenz post infectionem (2 bis 3 Wochen p.i.). Titer homologer Antikörper sind nunmehr nahezu 3 Jahre p.i. nachweisbar.
Die Darstellung eines dieser Fasciola-Antigene gelingt durch Kombination von Gelfiltration an Sephadex® G 75 und Polyacrylamid-Disc-Elektrophorese im präparativen Maßstab.
Das Präparat erscheint einheitlich im PAA-Disc-Elektropherogramm, im Präzipitationsverhalten gegen Totalhomogenatextrakt-Hyperimmunseren (Kaninchen) und Seren experimentell infizierter Kaninchen, in der Ultrazentrifugen-Sedimentation, nach isoelektrischer Fokussion und im Präzipitationsverhalten gegen homologes Antiserum (Maus).
Das Molekulargewicht des Antigens errechnet sich nach dem Geldiffusionsverhalten (Sephadex® G 75, fine) im Vergleich mit Human-Albumin, Trypsin und Ribonuklease mit 22,1 · 103, nach dem Sedimentationsverhalten in der Ultrazentrifuge mit 8,0 · 103 bis 12,0 · 103 (Pedersen, 1945).
Das Absorptionsmaximum liegt bei 260 nm.
Das Protein-Antigen weist einen Lipidanteil von ℞ 6% auf.
Das Mikrolipogramm zeigt Cholesterinester (27,1%), Triglycerid (24,6%), ein Steroid (5,6%), Cholesterin (9,4%), Monoglycerid (11,2%) und freie Fettsäuren (21,6%). Phospholipide und Fettsäureester wurden nicht festgestellt.
Die qualitative und quantitative Aminosäure-Analyse (außer Tryptophan) des Proteinanteils nach HCl-Hydrolyse führte bisher zum Nachweis von Ornithin (0,74% Aminosäure/mg Antigen), Lysin (10,7%), Histidin (3,24%), Arginin (3,36%), Asparaginsäure (7,63%), Threonin (4,76%), Serin (4,24%), Glutaminsäure (12,24%), Prolin (20,14%), Glycin (3,76%), Alanin (4,76%), Valin (6,84%), Methionin (2,31%), Isoleucin (3,86%), Leucin (5,25%), Tyrosin (2,44%) und Phenylalanin (3,68%).
Auf Grund der positiven PAs-Reaktion im PAA-Disc-Elektropherogramm kann mit einem Kohlenhydrat-Anteil des Antigens gerechnet werden.
Serologische Spezifitätsprüfungen gegen einige Trematoden-Hyperimmunseren (Fasciola gigantica, Schistosoma mansoni, S. japonicum, Paragonimus westermani, P. myasakii, P. ohirai, Clonorchis sinensis und Dicrocoelium dendriticum) weisen das Antigen in der Agarosegel-Diffusion als vermutlich Fasciola-gattungsspezifisch aus.
Prüfungen auf parasitenspezifische Ausprägung des Antigens deuten im Rahmen der durchgeführten Untersuchungen auf eine Fasciola-spezifische Prägung. Dem Präzipitatsystem scheint kein Autoimmunmechanismus zugrunde zu liegen.
Summary
In the present study the identification, purification and characterization of a precipitating „functional“ antigen from crude extract of adult Fasciola hepatica tissue are reported:
Immunoelectrophoretic analysis of crude extract revealed 5–7 precipitates as identified with sera from experimentally infected rabbits. Two of these precipitating extract components formed heavy long trailing precipitate bands soon after infection. First precipitate formation developed using sera collected 2–3 weeks p.i. These two precipitates could be observed for now approximately 3 years p.i.
One of these highly reactive precipitating extract elements was purified by means of gel filtration on Sephadex® G-75 columns and preparative polyacrylamide gel disc electrophoresis.
Purified antigen seems to be homogenous as analysed in polyacrylamide gel disc electrophoresis, ultracentrifuge patterns, after electrofocusing procedure and in immunoelectrophoresis using rabbit crude extract hyperimmune sera, sera from experimentally infected rabbits and homologous antisera from mice after repeated booster injections of purified antigen.
Molecular weight of antigen is estimated 8,0–22,1 · 103 as determined by Sephadex® G-75 (fine) column gel diffusion technique by comparison with human serum albumin, trypsin and ribonuclease and sedimentation during ultracentrifugation (s 20 w℞ 1,1), respectively (Pedersen, 1945). Absorption maximum was found at 260 nm.
The purified protein antigen was found to contain ℞ 6% lipid. Microlipograms revealed the presence of esterified cholesterol (27,1%), triglyceride (24,6%), unknown steroide (5,6%), non esterified cholesterol (9,4%), monoglyceride (11,2%) and free fatty acids (21,6%). No phospholipids and esterified fatty acids could be detected. Qualitative and quantitative amino acid analysis of HCl-hydrolysates were carried out: ornithine (0,74 per cent amino acid per mg antigen), lysine (10,7%), histidine (3,24%), arginine (3,36%), aspartic acid (7,63%), threonine (4,76%), serine (4,24%), glutamic acid (12,24%), proline (20,14%), glycine (3,76%), alanine (4,76%), valine (6,84%), methionine (2,31%), isoleucine (3,86%), leucine (5,25%), tyrosine (2,44%) and phenylalanine (3,68%). The antigen may have some protein-bound carbohydrate as indicated by positive PAS-reaction in polyacrylamide gel. Immunodiffusion of antigen and rabbit hyperimmune serum from adult Fasciola gigantica visualized one precipitate. No precipitate developed using hyperimmune sera from adult Schistosoma mansoni, S. japonicum, Paragonimus westermani, P. myasakii, P. ohirai, Clonorchis sinensis and Dicrocoelium dendriticum.
The purified antigen seems to be specific fluke material.
