Zusammenfassung
Imipramin, Dibenzepin, Desmethyl-Imipramin (Desipramin) und Desmethyl-Amitriptylin (Nortriptylin) potenzieren nach einer Vorbehandlungszeit von 30 min die 5-Hydroxytryptophan(5-HTP)-Hyperthermie des Kaninchens. Amitriptylin besitzt bei 30 minütiger Vorbehandlung eine schwache Hemmwirkung, potenziert dagegen das 5-HTP-Fieber nach einer Latenzzeit von 18 Std. Thioridazin vermag weder bei einer 1/2- noch einer 18 stündigen Vorbehandlungszeit 5-HTP zu potenzieren, im Gegenteil vermindert es die 5-HTP-Wirkung. Da 5-HTP zu einem Anstieg des Serotonins im Zentralnervensystem führt, und da das 5-HTP Fieber mit Monoaminoxydasehemmern verstärkbar, mit Serotoninantagonisten hemmbar ist, kann man annehmen, daß die zentrale Serotoninvermehrung in einem ursächlichen Zusammenhang mit der durch 5-HTP ausgelösten Temperatursteigerung steht. Daß die tricyclischen Antidepressiva das 5-HTP Fieber potenzieren, könnte demnach als eine Interferenz mit zentralen Serotoninwirkungen verstanden werden. Dabei deuten die geschilderten Befunde darauf hin, daß die Dimethyl-Verbindungen Imipramin und Dibenzepin direkt zu einer 5-HTP-Potenzierung führen, während Amitriptylin erst nach einer Metabolisierung im Organismus in seine aktive Form übergeht.
Summary
The tricyclic antidepressants imipramine, dibenzepine, desipramine and nortriptyline are shown to potentiate the fever induced by 5-Hydroxytryptophan (5-HTP) in the rabbit. They do so after a pretreatment time of thirty minutes where as amitriptyline possesses no potentiating effect after the same pretreatment period but gives a very clear potentiation of 5-HTP fever eighteen hours after injection. Thioridazine cannot be shown to potentiate the 5-HTP fever nor after a half nor after eighteen hours.
Because 5-HTP increases the serotonin-content of the central nervous system and because 5-HTP fever can be reinforced by monoaminoxydase-inhibitors and is reduced by antagonists of serotonin, one may assume that there is a causal relationship between the increase of serotonin in the animal's brain and the fever induced by 5-HTP. It is suggested that potentiation of 5-HTP fever by the antidepressants is a sign of interference of these compounds with brain serotonin. The findings reported suggest that a central potentiation of serotonin can be achieved by the N-dimethyl compounds imipramine and dibenzepine, whereas amitriptyline be metabolized into a active derivative, perhaps the desmethylated congener.
