Bei gesunden Ratten wurden 21 freie Plasmaaminosäuren nebeneinander quantitativ bestimmt und daraus der freie Gesamtaminostickstoff berechnet. Abgesehen von Glykokoll und Glutamin stimmen die Werte gut mit früher für den Menschen ermittelten Werten überein.

Bei anderen Ratten wurde mit hohen CCl4-Dosen eine akute, schwere Leberschädigung gesetzt. Die Tiere wurden 24 Std, 48 Std, 14 Tage oder 23 Tage später getötet und die freien Plasmaaminosäuren quantitativ bestimmt. Bei einigen Tieren wurden während der ersten Tage auch die Harnaminosäuren untersucht. Die Lebern sämtlicher Ratten wurden histologisch aufgearbeitet.

Den verschiedenen Vergiftungsstadien entsprachen jeweils charakteristische Veränderungen der freien Plasmaaminosäuren. Zu Beginn stieg die Konzentration der meisten Aminosäuren außer Glutaminsäure und Arginin steil an. Der Gesamtmino-N betrug 200% der Norm. Später verhielten sich die einzelnen Stoffe sehr unterschiedlich. Dadurch wurden ihre physiologischen Relationen zueinander stark verschoben und es resultierten erhebliche Imbalanzen im Aminosäurespektrum.

Da sich die Konzentrationsänderungen der Einzelaminosäuren gegenseitig kompensieren, war der Gesamtminostickstoff bereits nach 48 Std nicht mehr signifikant erhöht und stieg auch im Spätstadium nicht mehr wesentlich an. Er unterliegt also auch bei schwerer toxischer Leberschädigung einer strengeren Regulation als seine einzelnen Komponenten.
