Zusammenfassung
Die kontrahierende Wirkung von Phosphatidsäure auf die Taenia coli wurde bei verschiedenen Konzentrationen von Na+, K+ und Ca++ studiert. Alle 3 Ionenarten sind zur Auslösung von Kontraktionen notwendig.
Calcium ist für die Phosphatidsäurewirkung besonders wesentlich. Bei vermindertem Ca-Gehalt und häufiger Anwendung in kurzen Zeitabständen zeigt Phosphatidsäure ein Tachyphylaxiephänomen. In calciumfreier Lösung wird der Muskel langsam, bei Zugabe von EDTA schnell gegenüber Phosphatidsäure unempfindlich.
Die Befunde werden durch die Annahme erklärt, daß Phosphatidsäure mit Calcium reagiert, das in der Membran der Muskelzelle gebunden ist. Dadurch wird der Membranerregungsprozeß ausgelöst, dem die Kontraktion folgt. Für die erregende Wirkung von Phosphatidsäure durch direktes Einbringen von Calcium als lipoidlösliches Phosphatidat in die Zelle — unter Umgehung der Membranerregung — fand sich kein Anhaltspunkt.
Acetylcholin ist weniger stark von Ca-Ionen abhängig als Phosphatidsäure.
Summary
The contracting effect of phosphatidic acid on isolated strips of guinea pig taenia coli has been studied using various concentrations of Na+, K+ and Ca++. No contraction is produced if one of these ions is omitted from the bath-fluid.
Phosphatidic acid seems to be effective primarily by interaction with Ca++. In Ca-deficient Tyrode solution, the taenia becomes tachyphylactic to the action of phosphatidic acid. In the absence of calcium the muscle slowly becomes unresponsive, or more quickly if EDTA is added.
The findings are explained by assuming that phosphatidic acid combines with Ca++ bound at the cell surface. This triggers membrane excitation which is followed by contraction. No results have been obtained which would indicate that phosphatidic acid acts by directly transporting Ca++ into the cell, as a lipidsoluble salt, thereby inducing contraction without previous membrane excitation.
Acetylcholine is less dependent on Ca++, than phosphatidic acid.
