Zusammenfassung
Die parenterale Verabfolgung von Na2Ca-ÄDTA und Na3Ca-DTPA bewirkt innerhalb weniger Stunden eine Hemmung der alkalischen Phosphatase in Prostata, Serum und in besonders starkem Maße in der Nierenrinde. Die Dauer der Hemmung kann in Abhängigkeit von der Dosis bis zu 4 Tagen betragen. Das gehemmte Enzym wird in vitro durch Zn2+ reaktiviert. Na3Ca-DTPA ist in vivo zweimal, in vitro 100 mal wirksamer als Na2Ca-ÄDTA, während Na3Zn-DTPA die alkalische Phosphatase überhaupt nicht inaktiviert. Die Befunde sprechen dafür, daß es sich bei der Inaktivierung des Enzyms um einen primären Effekt, d.h. um eine Bindung des Zn der alkalischen Phosphatase durch die Chelatbildner, handelt. Obgleich in quantitativer Hinsicht eine enge Korrelation zwischen der Inaktivierung der alkalischen Phosphatase und der allgemeinen und Nephrotoxicität der Chelatbildner besteht, kann die pathogenetische Bedeutung der Befunde noch nicht übersehen werden.
