Zusammenfassung
Der Einfluß von akuter und chronischer DDT-Vergiftung auf den Stoffwechsel der unveresterten Fettsäuren (UFS) wurde am isolierten epididymalen Rattenfettkörper in vitro geprüft. Außerdem wurde der UFS-Gehalt im Plasma und im Fettgewebe zur Beurteilung der Verhältnisse in vivo untersucht.
Die 14tägige Zufuhr von 200 ppm DDT im Futter hatte keinen Einfluß auf den Fettsäurestoffwechsel des Fettgewebes. Dagegen löste die akute DDT-Vergiftung (200 mg/kg oral) eine starke Mobilisation von UFS aus. Dieser Effekt beruhte nicht auf einer direkten peripheren Wirkung von DDT auf das Fettgewebe, sondern stand mit den zentralen Erregungserscheinungen in Zusammenhang. Dementsprechend ließen sich diese Stoffwechselveränderungen durch Urethannarkose aufheben. Weiterhin wurde die erhöhte Freisetzung von UFS durch Vorbehandlung mit dem β-Receptorenblocker Nethalide (40 mg/kg s.c.) vollständig verhindert. Als Ursache der DDT-Wirkung auf den Fettstoffwechsel kommt daher eine Stimulierung des sympathischen Nervensystems durch akut toxische Dosen in Betracht.
Summary
The metabolism of unesterified fatty acids (UFA) was studied in the isolated epididymal fat pad of rats poisoned by DDT. In addition the concentration of UFA in adipose tissue and in blood plasma was determined.
A diet containing 200 ppm DDT given for 14 days did not alter the metabolism of fatty acids in adipose tissue. In the course of an acute poisoning with an oral dose of 200 mg/kg however, a considerable mobilisation of UFA was observed. This effect was not caused by a direct peripheral action of DDT on adipose tissue. It was associated with the central nervous excitation which is the characteristic feature of acute DDT poisoning. Accordingly the metabolic changes were abolished by urethane anaesthesia. Nethalide (40 mg/kg s.c.), an adrenergic β-receptor blocking agent, completely blocked the mobilisation of UFA in poisoned animals. This suggests that a stimulation of sympathetic nervous system is the primary cause for the action of DDT on lipid metabolism.
