Summary
1. Like Reserpine the Rauwolfia alkaloids Canescine and Rescinnamine deplete the adrenaline stores of the adrenal medulla of rabbits. Rescinnamine the least sedative acting drug also causes the smallest depletion of the suprarenals.
2. Rabbits react with a more severe depletion of the adrenal medulla than rats and mice.
3. Other sedative drugs like Pentobarbitone, Urethane and Chlorpromazine do not change the adrenaline content of the adrenal gland.
4. In cats the percentage content of adrenaline and noradrenaline remaines unchanged irrespectively of the degree of depletion, while in rats and mice after the injection of Reserpine the percentage of adrenaline was found to be greater than that in the normal gland.
5. Experiments on rabbits in which the adrenal gland had been denervated lead to the following results:

a)
After denervation of one gland by cutting the splanchnic nerve a week previously Reserpine in a large dose (4 mg./kg.) depleted much more the intact gland than the denervated one. On the other hand the depletion of both glands was nearly equal after the administration of 0,1 or 0,25 mg./kg.

After section of the spinal cord at the level of C6 Reserpine was found to be ineffective.
6. In contrast to rabbits Reserpine caused a normal depletion of the adrenal gland after section of the spinal cord in rats.
7. The experiments were undertaken to find out whether the depletion of the adrenal medulla is due to a central or a peripheral action of Reserpine.
