Zusammenfassung
1. Die Bestimmung des ATP-, Calcium- und Magnesium-Gehaltes isolierter chromaffiner Granula ergab einen molaren Quotienten ATP/Ca+Mg von 1,21. Nach einer 30 und 60 min langen Inkubation bei 37° C ist die spontane Brenzcatechinamin- und ATP-Freisetzung von einer prozentual gleich starken Calcium- und Magnesium-Freisetzung begleitet, so daß der molare Quotient ATP/Ca+Mg unverändert bleibt. Nach 2 Std langer Inkubation sinkt der Quotient ab, da der Calcium-Gehalt nicht mehr proportional zu der ATP- und Magnesium-Freisetzung abnimmt. Während eine Inkubation bei 0° C werden weder Brenzcatechinamine und ATP noch Calcium und Magnesium freigesetzt.
2. Die Inkubation chromaffiner Granula mit RN-ase verursacht neben der RNS-, ATP- und Amin-Freisetzung auch eine Freisetzung prozentual gleichen Umfangs von Calcium und Magnesium, jedoch keine wesentliche Änderung des Eiweiß-Gehaltes der Granula. Die molaren Quotienten Amin/ATP und ATP/Ca+Mg bleiben unverändert.
3. Reserpin und Segontin vermögen aus den Granula Brenzcatechinamine, ATP, Calcium und Magnesium prozentual gleichstark freizusetzen. Der Eiweiß- und RNS-Gehalt der Granula sowie die molaren Quotienten Amin/ATP und ATP/Ca+Mg werden durch Reserpin bzw. Segontin nicht beeinflußt.
4. Die durch Calcium bei 37° C und bei 0° C bedingte ATP-Freisetzung geht mit einer Magnesium-Abgabe einher. Der Abgabe von 2 Mol ATP entspricht die Abgabe von 1 Mol Magnesium. Der RNS-Gehalt der Granula bleibt unverändert.
5. Tyramin setzt aus den chromaffinen Granula Brenzcatechinamine, jedoch kein ATP, Calcium und Magnesium frei. Seine Wirkung ist von der Phosphat-Konzentration abhängig.
6. Durch osmotische Lyse der Granula werden freigesetzt: 100% des ATP, 94% der Brenzcatechinamine, 61% der RNS und des Calciums, 41% des Eiweißes und 48% des Magnesiums.
7. Segontin und Reserpin vermögen aus der isoliert durchströmten Nebenniere Brenzcatechinamine freizusetzen. Ihre Wirkung ist wie diejenige des Tyramins calcium-unabhängig.
Die Bedeutung der divalenten Ionen Calcium und Magnesium für die Speicherung der Nebennierenmark-Hormone in den chromaffinen Granula wird diskutiert.
Summary
1. Chromaffin granules were isolated from 1 g suprarenal medulla of cattle. They contain 4.17 μmoles ATP, 2.3 μmoles calcium and 1.14 μmoles magnesium. The molar ratio ATP/Ca+Mg is 1.21. The spontaneous release of catecholamines and ATP at 37° C (30 and 60 min) is accompanied by a release of calcium and magnesium to the same percentage degree so that the molar ratio ATP/Ca+Mg of the granules remains unchanged. After 2 hours incubation at 37° C the molar ratio decreases, as the release of calcium ceases in contrast to that of ATP and magnesium. At 0° C neither amines and ATP nor calcium and magnesium are released.
2. Incubation experiments with RN-ase (240 μg/ml) have shown that this enzyme increases the release of RNA, catecholamines, ATP, calcium and magnesium to the same percentage extent while the protein content of the granules remains constant.
3. Reserpine (0.2 μmoles/ml) and prenylamine (0.06 μmoles/ml) cause at 37° C a release of catecholamines, ATP, calcium and magnesium from the chromaffin granules. The protein and RNA content of the granules as well as the molar ratios amine/ATP and ATP/Ca+Mg remain unchanged.
4. Calcium (5 μmoles/ml) produces at 37° C and 0° C a concomitant release of ATP and magnesium; 2 moles ATP are released together with 1 mole magnesium.
5. Tyramine (10 μmoles/ml) releases only catecholamines. It has no effect on the ATP, calcium and magnesium content of the granules. The tyramine induced release of catecholamines is dependent on the phosphate concentration of the incubation medium.
6. Under osmotic lysis of the granules with distilled water (10 min, 0° C) are released: 100% of the ATP, 94% of the catecholamines, 61% of the RNA and of the calcium, 41% of the protein and 48% of the magnesium.
7. Experiments on isolated perfused suprarenals of cattle have shown that the injection of both reserpine (2.8 μmoles) and prenylamine (0.6 μmoles) releases catecholamines to about the same extent. Their action, as well as that of tyramine, is not dependent on the presence of calcium.
The role of both calcium and magnesium for the storage of catecholamines in the chromaffin granules is discussed.
