Zusammenfassung
An elektrisch gereizten linken Meerschweinchenvorhöfen wurde der Einfluß der β-Receptoren blockierenden Substanz 1-(3-Methylphenoxy)-2-hydroxy-3-isopropylamino-propan (MHIP; Kö 592) auf die Kontraktionskraft, die Refraktärzeit, auf intracellulär gemessene Aktionspotentiale und auf die positiv inotrope Adrenalinwirkung untersucht.
MHIP hatte am Meerschweinchenvorhof Wirkungen, die mit denen von Chinidin vergleichbar sind. Es wirkte negativ inotrop, verlängerte die Refraktärzeit, reduzierte die maximale Anstiegssteilheit des Aktionspotentials, verminderte das Umkehrpotential und verlängerte die Aktionspotentialdauer. Die negativ inotrope Wirkung von MHIP wurde durch Vorbehandlung der Tiere mit Reserpin fast vollständig aufgehoben. Durch die Vorbehandlung mit Reserpin sank der fluorometrisch bestimmte Noradrenalingehalt in den Herzkammern der Meerschweinchen auf weniger als 2% des Kontrollwertes.
Folgende Versuchsergebnisse sprechen gegen eine Abhängigkeit der β-adrenolytischen von den chinidinartigen Wirkungen am linken Meerschweinchenvorhof:

In einem Konzentrationsbereich von 5·10−8 bis 5·10−7 g/ml nahm die β-adrenolytische Wirkung von MHIP wesentlich stärker zu als die chinidinartigen Wirkungen.

Die negativ inotrope Wirkung von MHIP war in einem Konzentrationsbereich von 5·10−8 bis 5·10−7 g/ml nicht mit der β-adrenolytischen Wirkung korreliert. Ebenso war der Einfluß von 5·10−7 g/ml MHIP auf die Refraktärzeit nicht mit der β-adrenolytischen Wirkung korreliert.

5·10−7 g/ml MHIP hemmte die positiv inotrope Adrenalinwirkung deutlich mehr als eine äquimolare Konzentration von Pronethalol. Bei dieser Konzentration hatten MHIP und Pronethalol einen gleich starken Einfluß auf die Kontraktionskraft und auf die Refraktärzeit.
Chinidin hemmte in einem Konzentrationsbereich von 10−6 bis 5·10−5 g/ml die positiv inotrope Adrenalinwirkung nicht. Eine chinidinartige Wirkung hat also am Vorhof nicht zwangsläufig eine β-Receptoren-Blockade zur Folge.
Summary
A comparison was made of the actions of the beta adrenergic blocking agent 1-(3-methylphenoxy)-2-hydroxy-3-isopropylamino-propane (MHIP; Kö 592) and quinidine. The effects on the following parameters were studied on isolated left guinea-pig atria stimulated electrically: contractile force, refractory period, intracellularly recorded potentials and the positive inotropic action of adrenaline.
Some effects of MHIP were similar to those of quinidine. MHIP prolonged the refractory period and reduced the maximal rate of rise and the overshoot of the action potential. The duration of the action potential was prolonged. The force of contraction was decreased. The negative inotropic action of MHIP was nearly completely abolished by pretreatment of the guinea-pigs with reserpine. In the pretreated animals the noradrenaline concentration of the heart determined fluorometrically was lowered to less than 2% of the control values.
From the following results it is concluded that the beta adrenergic blocking action is not related to quinidine-like effects in the guinea-pig left atrium:

In concentrations ranging from 5·10−8 to 5·10−7 g/ml the dose-response curve of the beta adrenergic blocking action of MHIP had a much steeper slope than the dose-response curves of the quinidine-like effects.

There was no correlation of the negative inotropic effect of MHIP (5·10−8 to 5·10−7 g/ml) with its beta adrenergic blocking action. The same was true for the action of 5·10−7 g/ml MHIP on the refractory period and on the positive inotropic effect of adrenaline.

In equimolar concentrations MHIP was essentially more powerful in blocking beta receptors than pronethalol. However, the effects on refractory period and on contractile force of both compounds did not differ.
Quinidine (10−6 to 5·10−5 g/ml) did not block the positive inotropic actions of adrenaline. Thus in the isolated atrium a quinidine-like effect does not necessarily result in beta adrenergic blocking activity.
