Der beschleunigte Abbau von Pharmaka in den Lebermikrosomen unter dem Einfluß von Luminal

Summary

The microsomes of rats treated with Phenobarbital (Luminal) oxidize Hexobarbital (Evipan) and demethylate Methylaminoantipyrin (M. A. A.) with a much higher rate than microsomes of untreated rats. Correspondingly the duration of anaesthesia with Hexobarbital is significantly shortened in treated rats. One injection of 100 mg./kg. Phenobarbital s.c. is sufficient to accelerate the in vivo and in vitro oxidation of Hexobarbital and the in vitro demethylation of M. A. A. After a delay of about 12 hours the speed of oxidation rises, reaches its maximum after 2 days, and decreases in 5–7 days to normal values.

Comparing the microsomes of male and female rats those of the males have an oxidative activity 3–4 times higher.

The duration of Hexobarbital anaesthesia of rabbits and mice after just 1 injection of Phenobarbital is shortened in a similar manner. Comparably the concentration of Hexobarbital decreases in the blood of rabbits with a much higher rate, if these animals were treated with Phenobarbital.
