Zusammenfassung
NAD und NADP werden neben 3-APAD und 3-APADP in Hirnextrakten von Ratten mit einem spektrofluorometrischen Verfahren quantitativ bestimmt.
Die Analysen verschiedener Hirnregionen ergaben, daß der Umfang des Austausches von Nicotinsäureamid gegen 3-Acetylpyridin in die Pyridinnucleotide nicht gleich ist. Nach Verabreichung von 100 mg/kg 3-Acetylpyridin an weiße Ratten werden die höchsten Werte an 3-APADP im Hippocampus gefunden. Zwischen 44–48% der natürlichen Codehydrase II werden in das 3-Acetylpyridin-enthaltende Derivat umgewandelt. Die dadurch bedingte Verlangsamung des Wasserstofftransportes zwischen Substrat und NADP-bedürftigen Dehydrogenasen kann die Ursache der neurologischen Störungen sein.
Im Verlauf der Vergiftung mit 3-Acetylpyridin nimmt die Konzentration an oxydiertem NAD und NADP ab.
Summary
Using a spectrofluorometric method, the amounts of NAD and NADP as well as 3-APAD and 3-APADP found in white rat brain extracts were determined quantitatively.
An analysis of different regions of the brain showed that the extent of exchange between nicotinic acid-amide and 3-acetylpyridine in the pyridine nucleotide molecule varies according to region, the highest values of 3-APADP being found in the hippocampus. Given a dosis of 100 mg/kg 3-acetylpyridine, it was found that between 44–48% of the normal Codehydrase II had been transformed into the 3-acetylpyridine derivate. It is considered possible that the neurological symptoms seen by 3-acetylpyridine poisoning are a result of the decreased hydrogentransport between substrate and NADP-dependant dehydrogenase.
As a 3-acetylpyridine poisoning progresses, the amount of oxydized NAD and NADP are found to decrease.
