Zusammenfassung
Es wird eine spektrofluorometrische Methode zur quantitativen Bestimmung von 6-Aminonicotinsäureamid, 6-Aminonicotinsäureamid-adenin-dinucleotid (6-ANAD) und 6-Aminonicotinsäureamid-adenin-dinucleotid-phosphat (6-ANADP) beschrieben.
Bei der Vorreinigung der Extrakte durch Säulen-oder Papierchromatographie werden die besten Ergebnisse mit dem papierchromatographischen Verfahren erzielt.
In den Extrakten aus Rattennieren wird eine noch unbekannte Substanz mit einem charakteristischen Anregungsspektrum nachgewiesen, deren Elimination für die quantitative Bestimmung des oxydierten NADP notwendig ist.
6 Std nach der intraperitonealen Applikation von 6-Aminonicotinsäureamid werden 38,5% 6-Aminonicotinsäureamid-adenin-dinucleotid und bis zu 70% 6-Aminonicotinsäureamid-adenin-dinucleotid-phosphat in den Fraktionen der natürlichen Pyridinnucleotide aus Rattennieren gefunden. Die Folgen der Synthese pathologisch veränderter Pyridinnucleotide für den Stoffwechsel der Zelle werden diskutiert.
Summary
A spectrofluorometric method for the quantitative determination of 6-Amino-nicotinamide, 6-Amino-nicotinamide-adenine-dinucleotide (6-ANAD), and 6-Amino-nicotinamide-adenine-dinucleotide-phosphate (6-ANADP) is described.
Better results were obtained by pre-cleansing the extracts through paper-rather than column-chromatography.
Included in the rat kidney extracts is an unknown substance with a characteristic activating spectrum which must be eliminated by any quantitative determination of oxidated NADP.
Six hours after intra-peritoneal injection of 6-Aminonicotinamide, 38.5 per cent 6-Amino-nicotinamide-adenine-dinucleotide and as much as 70 per cent 6-Amino-nicotinamide-adenine-dinucleotide-phosphate were found in the fractions of naturally occuring Pyridin-nucleotides from rat kidneys. The possible consequences of the synthesis of pathologically changed Pyridin-nucleotides for the metabolism of the cell are discussed.
