Zusammenfassung
An Meerschweinchenpapillarmuskeln wird der Einfluß von Calcium bei 25 und bei 35° auf folgende Größen bestimmt: Kontraktionskraft (F), Anstiegszeit (t) und mittlere Anstiegssteilheit der isometrischen Kontraktion S (F/t).
Die Natriumkonzentration ist auf 70 mM (bei osmotischem Ausgleich durch Saccharose) vermindert, um maximale Calciumwirkungen zu ermöglichen. Die Calciumkonzentration wird zwischen 0,15 und 9,6 mM variiert.
Während die Anstiegszeit über den ganzen Calciumbereich eine annähernd gleiche negative Temperaturabhängigkeit aufweist, ist die Temperaturabhängigkeit von Kontraktionskraft und Anstiegssteilheit nur bei niedrigen Calciumkonzentrationen negativ, bei hohen jedoch positiv. Der Q 10 für die maximale Kontraktionskraft (bei 35°C und 4,8 mM Ca++; bei 25°C und 2,4 mM Ca++) übersteigt nicht den Wert 1. Der Q 10 für die maximale mittlere Anstiegssteilheit (bei 4,8 mM Ca++, entsprechend 19,2 mM Ca++ bei 140 mM Na+) beträgt 2,2.
Als ein Maß für den der Kontraktion zugrundeliegenden „Aktivitätszustand“ A wird der Wert S/t (bzw. F/t 2) betrachtet. Sein Temperaturkoeffizient wird bei maximaler Calciumwirkung (4,8 mM Ca++, 70 mM Na+) mit 3,6 bestimmt.
Summary
The influence of calcium on contractile force (F), peak time (t) and rising steepness S (F/t) of the isometric contraction of the guinea pig papillary muscle has been investigated at 25 and 35°C.
The sodium concentration was decreased to 70 mM (with an osmotically equivalent addition of sucrose) to render possible a maximal calcium effect. The calcium concentration was varied between 0.15 and 9.6 mM.
Whereas the peak time shows a negative temperature dependence over the whole calcium range, the temperature dependence of contractile force and rising steepness is negative only at low but positive at high Caconcentrations.
The temperature coefficient, Q 10, for maximal force of contraction (at 35°C and 4.8 mM Ca++; at 25°C and 2.4 mM Ca++) does not exceed 1.
The rising steepness has a temperature coefficient, Q 10, of 2.2 at 4.8 mM Ca++ and 70 mM Na+ (corresponding to 19.2 mM Ca++ at 140 mM Na+).
The value S/t (or F/t 2) has been regarded as a magnitude of the “state of activity” of the contracting muscle. Its temperature coefficient at the maximal Ca-effect has been estimated with 3.6.
