Zusammenfassung
An narkotisierten, künstlich beatmeten Ratten wurde die Wirkung von Nicotin bei Injektion in den 4. Hirnventrikel und bei Applikation an der Area ventrolateralis der Medulla untersucht. Dabei ergab sich:

Injektionen von 1–3 μg Nicotin in den 4. Ventrikel lösten einen Atmungsstillstand und eine Dauererregung im efferenten Vagus aus, die der Reaktion auf 1,0 mg Nicotin bei i.v. Zufuhr entspricht. Tiere, die durch i.v. Nicotin-Injektionen tachyphylaktisch gemacht waren, reagierten auch auf intraventrikuläre Nicotingaben nicht mehr.

Hexamethonium verhinderte bei Injektion in den 4. Ventrikel (150 μg) die Apnoe und die Vaguserregung nach i.v. Nicotin-Injektion vollständig. Auch nach Procain (50–100 μg) ließen sich ähliche antagonistische Wirkungen aufzeigen.

An der Area ventrolateralis der Medulla löste die lokale Nicotin-applikation 1–10 μg nur atmungssteigernde Effekte aus.
Die Versuche lassen vermuten, daß die zentralen Wirkungen hoher Nicotindosen, insbesondere Apnoe und Vaguserregung, bei der Ratte vornehmlich durch einen Angriffspunkt in der Umgebung des 4. Ventrikels zustande kommen. Die dort lokalisierten Receptorelemente zeigen eine pharmakologische Beeinflußbarkeit, die auf eine Ähnlichkeit mit den N-Receptoren in den vegetativen Ganglien hindeutet.
Summary
The effect of nicotine on the electrical activity of the efferent vagus and phrenic nerves has been investigated in anaesthetised rats under artificial respiration. The drug was injected into the fourth ventricle or applied topically into the subarachnoid space to the ventrolateral surface of the medulla.
The results were as follows:

Injections of 1–3 μg nicotine into the fourth ventricle produced respiratory standstill and an excessive excitation in the efferent vagus nerve. A similar reaction is obtained when 1,0 mg/kg nicotine is injected intravenously. Animals made tachyphylactic to nicotine given intravenously also failed to respond to nicotine when injected into the fourth ventricle.

The apnoe and stimulation of the vagus nerve produced by intravenous nicotine was completely abolished by an injection of hexamethonium (150 μg) into the fourth ventricle. Also procain (50–100 μg) produced a similar antagonistic effect.

When applied topically to the area ventrolateralis of the medulla 1–10 μg nicotine elicited a marked respiratory stimulation.
These results indicate that in the rat the “central” effects of high doses of nicotine as apnoe and stimulation of the vagus nerve are elicited from a site adjacent to the fourth ventricle. The pharmacological properties of the receptor elements there are similar to the N-receptors of vegetative ganglia.
