Zusammenfassung
1. Aus der sogenannten Kernfraktion von Rattennierenrinden-Homogenaten lassen sich durch nicht-kontinuierliche oder Schichten gradienten-Zentrifugation in Saccharose Basalmembranen anreichern.
2. Im elektronenmikroskopischen Bild stellen sich die Basalmembranen als elektronendichte Bänder dar, die bei hoher Vergrößerung 40 bis 60 Å dicke, netzartig verflochtene Filamente und ein interfilamentäres Matrixmaterial erkennen lassen.
3. Durch Ultraschalleinwirkung tritt an vielen Stellen eine Spaltung der Basalmembranen in zwei Lamellen ein.
4. Die chemische Aufarbeitung ergibt einen hohen Hydroxyprolingehalt der Basalmembranfraktion, der auf einen hohen Kollagenanteil in der Basalmembran der Niere hinweist.
Summary
1. Basement membranes could be enriched from the so-called nuclear fraction of rat kidney cortex homogenates by means of non-continuous or layer gradient centrifugation with sucrose.
2. The basement membranes appear in the electron micrograph as electron-dense bands which upon higher magnification exhibit network-like interlaced filaments of a thickness of 40–60 Å and an interfilamentary matrix material.
3. By application of ultra sound waves a cleavage of the basement membrane into two laminae is seen in many places.
4. Chemical analysis of this basement membrane fraction reveals a high content of hydroxyproline, giving evidence for the existence of a high collagen content in renal basement membranes.
