Zusammenfassung
1. Das Kation Serotonin wird durch die Proteine des menschlichen Serums entsprechend dem Massenwirkungsgesetz gebunden. Ein alkalisches Milieu begünstigt die Komplexbildung.
2. Die für Humanseren gemessene Serotoninbindung läßt sich auf die Albuminkomponente dieser Seren beziehen.
3. Ein Molekül Humanalbumin kann bei ph 8,8 in 0,05 m Tris-puffer maximal 11 Serotoninmoleküle binden. Die durchschnittliche Assoziationskonstante für den Albumin-Serotonin-Komplex wird mit k=6,5 · 102 l/Mol errechnet, die freie Energie der Bindung beträgt − ΔF°=3,52 kcal · mol−1.
Erhöhung der H+-Ionenkonzentration, Zn++- und Cu++-Ionen hemmen die Komplexbildung zwischen Albumin und Serotonin.
4. Die Hemmung der Serotoninwirkung durch humanes Serumalbumin im pharmakologischen Test liegt vielfach höher als die Serotoninbindung durch Albumin. Für die Hemmung der pharmakologischen Serotoninwirkung durch humanes Serumalbumin fand sich in unserer Versuchsanordnung eine logarithmische Abhängigkeit von der Zahl der gebildeten Albumin-Serotonin-Komplexe.
