Zusammenfassung
Die Untersuchung von Linol-, Linolen- und Arachidonsäure am Meerschweinchenileum, Kaninchenjejunum, Hamstercolon und Kaninchenblutdruck ergab, daß keine dieser mehrfach ungesättigten Fettsäuren eine darmkontrahierende bzw. blutdrucksenkende Wirkung hat. Erst wenn Gelegenheit zur Autoxydation der Fettsäurelösungen gegeben ist, werden sie wirksam. Behandlung mit Hämoglobin, Wasserstoffperoxyd oder Lipoxydase hat den gleichen Effekt wie Autoxydation.
Die darmerregende und blutdrucksenkende Wirkung der oxydierten Fettsäurelösungen beruht auf Fettsäurehydroperoxyden. Ascorbinsäure, FeCl2 oder Triphenylphosphin zerstören die Wirkung.
Hydroxyfettsäuren, wie Prostaglandin und wahrscheinlich Irin, haben eine von Peroxydbildung unabhängige Eigenwirkung. Wenn diese sehr schwach ist, können Hydroxyfettsäuren außerdem auch durch einen Peroxydgehalt darmkontrahierend wirken, wie am Beispiel der Ricinolsäure gezeigt wird.
Das Hamstercolon ist besonders empfindlich gegenüber der Eigenwirkung von Hydroxyfettsäuren und reagiert nur schwach auf Hydroperoxyde, während der Meerschweinchendarm besonders stark auf Hydroperoxyde anspricht. Die Verwendung dieser beiden Testobjekte und die Benutzung von Triphenylphosphin zur selektiven Peroxydzerstörung werden als Mittel zur Unterscheidung von Peroxyd- und Hydroxyfettsäurewirkungen unbekannter Extrakte vorgeschlagen.
Summary
Linoleic, linolenic, and arachidonic acid were assayed on isolated strips of guinea pig ileum, rabbit jejunum and hamster colon as well as on the blood pressure of rabbits. None of these polyunsaturated fatty acids had any gut contracting or hypotensive effect. However, the fatty acid solutions became active after they were allowed to autoxidize. Treatment of fatty acid solutions with haemoglobin, hydrogen peroxide or lipoxidase also led to pharmacologically active fatty acid preparations.
The intestine stimulating and hypotensive effect of the fatty acid solutions is due to hydroperoxides formed. Ascorbic acid, FeCl2 or triphenylphosphine abolish the activity.
Hydroxy-fatty acids, such as prostaglandin and presumably irin, have a stimulating effect, themselves, which is independent of formation of hydroperoxides. In addition, hydroxy-fatty acids can produce contraction of smooth muscle due to peroxide formation, as is shown to be the case with ricinoleic acid.
The hamster colon is highly sensitive to the direct action of hydroxy-fatty acids and is relatively insensitive to peroxides. In contrast, the guinea pig ileum responds particularly strongly to such peroxides. The use of these two intestinal preparations and of triphenylphosphine as a peroxide destroying agent is suggested as decisive tools for the identification of actions of hydroxy acids or hydroperoxides in unknown solutions.
