Zusammenfassung
Mit intracellulären Mikroelektroden wurden von Vorhof- und Kammermuskulatur von Meerscheinchenherzen Membran- und Aktionspotentiale abgeleitet. Die akute Wirkung von Trijodthyronin (T3 10−6 und 10−5 g/ml) auf diese elektrophysiologischen Meßgrößen wurde untersucht; außerdem wurden Veränderungen der intracellulären K- und Na-Konzentrationen in vitro und in vivo gemessen.
Die 90%ige Repolarisationsphase des Vorhofaktionspotentials wurde bei akuter Einwirkung einer T3-Konzentration von 10−6 g/ml verlängert, das Membranpotential stieg um 9 mV an. Membran- und Aktionspotential der rechten Kammer blieben bei dieser T3-Konzentration unverändert. Bei einer zehnfach höheren T3-Konzentration wurden die 20 und 50%igen Repolarisationsphasen des Vorhofaktionspotentials verkürzt, die 90%ige verlängert, das Membranpotential stieg nur geringfügig an. Die Repolarisation des Kammeraktionspotentials wurde in allen Phasen verlängert, das Membranpotential stieg deutlich an.
Bestimmungen der intracellulären K- und Na-Konzentrationen ergaben einen signifikanten Anstieg der Intracellulären K-Konzentration in 2 Std lang akut mit T3 (10−5 g/ml) behandelten Vorhofpräparaten. Vorhöfe stark thyreotoxischer Meerschweinchen wiesen ebenfalls gegenüber Normaltieren erhöhte K-Konzentrationen auf.
Die Befunde sprechen fűr eine Beeinflussung des Ionenhaushalts durch Schilddrüsenhormone. Die Refraktärperiode des Vorhofs wird verkürzt und dadurch die Flimmerbereitschaft gesteigert.
Summary
Intracellular membrane and action potentials were recorded from electrically driven auricles and ventricles of guinea pigs. The acute action of triiodothyronine (T3 10−6 and 10−5 g/ml) upon these electrophysiological properties was investigated. Changes in potassium and sodium concentrations were also measured in vitro and in vivo.
There was a marked prolongation of the 90% repolarisation phase and an increase of membrane potential during the acute action of T3 10−6 g/ml in auricles. Membrane and action potentials of the right ventricle were unchanged at this T3-concentration. Higher T3-concentrations (10−5 g/ml) shortened the 20- and 50% repolarisation phases but prolonged the 90% repolarisation phase of the auricular action potential. There was only a slight increase of membrane potential. In the ventricle all the repolarisation phases of the action potential were prolonged, and membrane potential was markedly increased.
The acute action of T3 10−5 g/ml in vitro produced an increased intracellular potassium concentration in auricles, and an increased potassium concentration was also observed in atria of thyrotoxic guinea pigs in vivo.
The results presented here indicate that thyroid hermones can influence the ionic distribution in the guinea pig heart, the refractory period being shortened, and fibrillation therefore occuring more readily.
