Zusammenfassung
Das Ausmaß der saluretischen Wirkung eines Präparates steht nicht in direktem Zusammenhang mit seinem in vitro gemessenen Hemmeffekt gegenüber der Carboanhydratase.
Saluretica können jedoch intermediär zu sehr aktiven Hemmstoffen dieses Ferments abgebaut werden.
Zudem kann auf Grund von Untersuchungen über die Kinetik der Carboanhydratase wahrscheinlich gemacht werden, daß auch Inhibitoren, die in vitro schwächer wirken als Acetazolamid, bei diuretisch wirksamer Dosierung eine Blockierung der Nierencarboanhydratase ergeben.
Eine Hemmung dieses Ferments kann auch durch direkte Aktivitätsmessung bei mit Saluretica behandelten Tieren beobachtet werden, und zwar tritt die Hemmung bei den verschiedenen Präparaten im gleichen Dosenbereich auf, der auch saluretisch wirksam ist.
Es darf aus diesen Gründen nicht ausgeschlossen werden, daß die Beeinflussung der Carboanhydratase eine bedeutende Rolle im Wirkungsmechanismus der Saluretica spielt.
Die charakteristische Wirkungsweise der Saluretica kann im Zusammenhang stehen mit der selektiven Passage und Lokalisation der Präparate in den verschiedenen Abschnitten der Tubuli.
