Zusammenfassung
Untersuchungen über die additive Wirkung verschiedener Herzglykoside bei i.v. Dauerinfusion an Meerschweinchen führten zu folgenden Ergebnissen:

Unterbricht man die Infusion nach 65–75% der tödlichen Dosis und füllt sofort anschließend mit g-Strophanthin auf, so ergibt sich mit Lanadoxin eine überadditive, mit Gitaloxin und Digitoxin eine additive Wirkung, mit Dt 21 und Helveticosid ein relativer und mit Gitaloxigenin ein absoluter Antagonismus. Infundiert man die Glykoside in umgekehrter Reihenfolge, so verschwinden sowohl die Potenzierung als auch der Antagonismus; unter Umständen werden beide in ihr Gegenteil verkehrt. Es wird versucht, diese Ergebnisse durch eine unterschiedliche Reversibilität der Bindung der Herzglykoside und einen kompetitiven Antagonismus zwischen Herzglykosiden und K+ zu erklären.

Nach einer Unterbrechung der Infusion während 2 Std war die Wirkung von Helveticosid, Lanadoxin, Gitaloxin und Digitoxin unverändert nachweisbar, wenn dasselbe Glykosid zur Nachtitration benutzt wurde. Trotzdem entwickelte sich nach Vorbehandlung mit Digitoxin eine Unterempfindlichkeit gegenüber g-Strophanthin, was auf die Bildung von Stoffwechselprodukten hinweist.

The additive effect of cardiac glycosides was investigated by intravenous infusion in guinea pigs.

In a first series of experiments, the infusion was interrupted after 65–75% of the lethal dose and immediately continued with ouabain until cardiac arrest occurred. An overadditive effect was observed with lanadoxin, an additive effect with gitaloxin and digitoxin, a relative antagonism with Dt 21 and helveticosid and an absolute antagonism with gitaloxigenin. Potentiation and antagonism disappeared when the sequence of the glycosides was reversed. The results are explained by a different reversibility of the combination between glycosides and receptor and by a competitive antagonism between glycosides and K+.

When the infusion of the glycosides was interrupted for two hours the effects of helveticosid, lanadoxin, gitaloxin and digitoxin were found undiminished, provided the same glycoside was used for the second infusion. However, an antagonism against ouabain developed after pre-treatment with digitoxin which can be accounted for by the formation of metabolic products.
