Die Wirkung der Nicotinsäure und anderer gefäßerweiternder Substanzen auf die Elimination von Kongorot

Summary

The influence of nicotinic acid (NA) and of other vasodilatory substances on the elimination of Congo red (CR) was studied in rabbits and dogs. The following results were obtained:

  1. 1.

    In rabbits the elimination constant rose with decreasing dose; the product k e · dose remained constant within a dose range from 5 to 20 mg/kg Congo red. In the same CR dose range NA in doses of 50 and 250 mg/kg produced a significant increase of the elimination rate of CR. There was a significant dose effect relation within a range of 5 to 250 mg/kg NA i.v.

  2. 2.

    In dogs NA, “Ronicol”, “Persantin”, “Duvadilan”, Papaverin and sodium nitrite produced a strong and dose depending rise in the elimination rate of 20 mg/kg CR i.v. The results were expressed in terms of relativ changes of CR total clearance and gave linear log dose effect relations for all the investigated vasodilatory substances. There was a correlation between the relative potency of the vasodilatory action and of the effect on the CR elimination within this series of vasodilators.

    The excretion of CR in the bile was significantely rised by the i.v. administration of 50 mg/kg NA. No measureable CR renal excretion could be found neither before nor after the injection of nicotinic acid.

The results were interpreted by an increase in capillary permeability to protein-dye complexes under the action of vasodilatory substances.
