Summary

The influence of a synthetic oxytocin preparation and of prednisolone hemisuccinate on the renal excretions of water, sodium, potassium and chloride and on the clearance of endogenous true creatinine was studied in 18 dogs anesthetized with hexobarbital and barbital. The animals were deprived of food and water for 12 hours before the experiments.

The intravenous injection of 100 mU/kg of oxytocin, but not of smaller doses, provoked a considerable increase in diuresis and sodium, chloride and potassium excretions. This increase lasted for approximately 1 hour. There was a simultaneous slight increase of the clearance of endogenous creatinine.

I.v. prednisolone hemisuccinate (3 mg/kg) had nearly the same effect on renal water and salt excretion in thirsty dogs as oxytocin. The diuretic effect of prednisolone, however, was not accompanied by an increase in the clearance of endogenous creatinine.

The diuretic and saluretic effects of oxytocin were not depressed, when a second injection was given 4 hours after the first.

The renal effects of oxytocin were completely suppressed by a preceding injection of prednisolone hemisuccinate. Similarily, prednisolone hemisuccinate had no diuretic or saluretic effects when injected after oxytocin.
