Summary
This study was precipitated by a patient suffering from progressive ataxia and spastic paralysis of the legs for several years with neither impairment of the cranial nerves nor any sensory troubles. His CSF showed normal cell counts on repeated spinal taps, however, protein levels were increased. One litter of newborn mice was inoculated intracerebrally with spinal fluid of the first tap; three months later five males of this litter developed ataxia, paralysis, and epileptic attacks. The histological evaluation of the animals revealed a condition similar to Scrapie with an extensive increase of glial cells and with a foam-like distruction of motor cells particularly in the anterior horn and brain stem. An outstanding feature was the formation of round bodies within these vacuoles, displaying a structure resembling a ball of wool. This material was also found at tissue sites where the ganglion cells had already completely disappeared. These experiments were performed on more than 400 animals during a period of 2 years. Histological studies of the normal female offspring of the inoculated litters which did not develop any symptoms, as well as of the respective mothers showed the same features in most of them, though to a lesser extent. Similar changes of the same extent were also found in animals not inoculated and killed immediately upon arrival from commercial sources. Nevertheless only the male animals died from this disease when they were inoculated either with three different specimens of spinal fluid obtained at different dates or with material passed through a filter not permeable to bacteria. The incubation period, however, was prolonged in the latter experiments. The results are compared with those of P. Pálsson, Field and Patterson who found similar phenomena in sheep and mice inoculated with material from M. S. patients. We have come to the conclusion that there is an epizootic disease of mice which resembles Scrapie, with a low morbidity and mortality and detectable only in NMRI mice supplied by german animal breeders. In our experiments the inoculation of spinal fluid seems to have provoked this epizootic disease to take a fatal course.
Zusammenfassung
Ausgangspunkt der Untersuchung war eine Patientin, die seit mehreren Jahren an einer langsam zunehmenden Ataxie und spastischen Lähmung der Beine litt, ohne Beeinträchtigung der Hirnnerven und ohne Sensibilitätsstörungen. Bei wiederholten Punktionen fanden sich normale Zellzahlen bei hohen Eiweißwerten. Der Liquor dieser Patientin wurde auf einen Wurf von Mäusen intracerebral verimpft. 5 Männchen dieses Wurfes bekamen eine ataktische Lähmung der Beine und epileptische Anfälle nach 3 Monaten. Die histologische Untersuchung ergab einen Befund, der an Scrapie erinnert mit ausgedehnter Vermehrung der Gliazellen und einer schaumartigen Entartung der großen motorischen Zellen der Vorderhörner des Rückenmarkes und des Hirnstammes. Ein besonderes Kennzeichen war die Bildung von runden Einschlußkörperchen innerhalb dieser Vacuolen mit der Struktur eines Wollknäuels. Dasselbe Material fand sich auch frei im Gewebe, wenn die Ganglienzellen schon völlig verschwunden waren.
Die histologische Untersuchung der weiblichen verimpften Tiere, die klinisch keine Erscheinungen hatten, und der Mütter der Tiere zeigte bei den meisten dieselben Veränderungen, obwohl in geringerer Ausprägung.
In einem gewissen Umfange konnten solche Veränderungen auch bei nicht beimpften Tieren gefunden werden, die unmittelbar nach der Einlieferung in unsere Ställe getötet worden waren. Indessen starben nur die männlichen Tiere an dieser Krankheit nach der Beimpfung mit drei verschiedenen Liquorproben, die zu verschiedenen Zeiten entnommen worden waren, und ebenso, wenn die Tiere mit Material beimpft worden waren, das über andere Saugmäuse und durch mikrobendichte Filter passiert worden war, Indessen verlängerte sich die Inkubationszeit in späteren Versuchen. Im Laufe von 2 Jahren haben wir die Untersuchungsbefunde von 400 Mäusen überprüft und bestätigt. Die Versuche wurden mit den Ergebnissen von Pálsson, Field u. Patterson verglichen, die ähnliche Beobachtungen bei der Verimpfung von Multiple Sklerose-Material auf Schafe und Mäuse gemacht hatten.
Wir schließen aus unseren Versuchen, daß es eine epizootische Mäusekrankheit gibt, welche der Scrapie ähnlich sieht, die eine geringe Morbidität und Mortalität hat und die wir nur bei NMRI-Mäusen aus deutschen Zuchtanstalten feststellen konnten. Bei unseren Versuchen hat aber die Verimpfung von Liquor einer Patientin offensichtlich dazu geführt, daß diese epizootische Krankheit einen tödlichen Verlauf nimmt.
