Detection of glycosphingolipid antigens ABH and Lewis on thin-layer plates using the PAP-method

Summary

Stroma from hemolyzed erythrocytes of blood groups 0, A1, B and A1B were obtained and subjected to butano/phosphate buffer extraction. This extract was separated using HPTLC, and the ABH and Le substances were detected on the chromatogram using the PAP technique. The staining of the bands allowed specific demonstration of the serologically active glycosphingolipids present in the ABH and Le blood group substances. The antigens of the ABO system showed a 3- to 12-band pattern. Each of the antigens Lea and Leb presented 3 bands. The slight differences in the levels of glycosphingolipids of equal chain lengths are probably due to differences in their chemical structures.

Zusammenfassung

Aus hämolysierten Erythrozyten der Blutgruppe 0, A, B, und A1B wurde die Membranmasse gewonnen und einer Butanol-Phosphatpufferextraktion unterzogen. Die so gewonnenen Extrakte wurden mittels HPTLC aufgetrennt. An den Chromatogrammen wurde der ABH- und Le-Nachweis mittels PAP-Technik durchgeführt. Die hierbei erzielte Bandenanfärbung erlaubt die spezifische Darstellung der serologisch aktiven Glykosphingolipide der ABH- und Le-Blutgruppensubstanzen. Die Antigene des ABO-Systems zeigen sich in Gestalt von 3- bis 12-bandigen Mustern. Die Antigene Lea und Leb weisen jeweils 3 Banden auf. Kleine Verschiebungen in den Höhenniveaus der Glykosphingolipidgruppen der gleichen Kettenlänge des ABO-Systems können auf Unterschiede im chemischen Antigenaufbau zurückgeführt werden.
