Summary
A quantitative colorimetric method to measure lignin content in wood has been developed. Red colored complex formed in presence of phloroglucinol-HCl was solubilised by addition of acetic acetyl bromide. Linearity was demonstrated between the absorption values at 545 nm of this complex and 0.5 to 2 mg of lignin of spruce or poplar wood, in presence or absence of ligninolytic fungi.
