Construction of a vector for probing the effect of co-expression of dnaY and secB upon secreted gene products in Escherichia coli

Summary

The agg/aga tRNA gene, dnaY, and the chaperonin gene secB have been combined on the same plasmid, pHB2. Therefore, the effect of increased secB expression upon accumulation of secreted proteins may be assessed, even for genes containing a high proportion of rare arginine codons. The effect of co-transformation of pHB2 upon yields of BPTI in Escherichia coli is discussed.
