Diversity and Antimicrobial Activity of Plant Associated Bacteria from Selected Medicinal Plants in Kutch, Dhinodhar Hill, Gujarat

A total of 19 plant associated bacteria were isolated through surface sterilization from three medicinal plants Salvodora oleoides, Commiphora wightii and Grewia tenax collected from the Kutch, Dhinodhar hill of Gujarat. A total of 190 organic solvent fractions were screened for antimicrobial activity by the agar well diffusion method; only six bacterial isolates (LGTKD4, SSOKD16, LSOKD6, SGTKD12, SSOKD1 and LSOKD9) showed a clearing zone around Escherichia coli, Proteus vulgaris, Staphylococcus aureus and Shigella flexneri. Maximum antimicrobial activity was observed with ethanol and methanol extracts and least with ethyl acetate. The TLC result showed that the compound more closely related to Kanamycin. The 16S rRNA gene sequencing results showed that the isolates LGTKD-4, SSOKD-16, LSOKD-6, SGTKD-12 and LSOKD-9 were Bacillus safensis and the isolates SSOKD-1 and LSOKD-6 belonged to Bacillus subtilis by comparing with the 16S rRNA database in NCBI, by BLAST analysis. This the first report of Bacillus safensis as plant associated bacteria with medicinal plant with antimicrobial activity.
