Abstract

Numerous applications in biotechnology would not be feasible without optimized enzymes. While changing the substrate specificity, enzyme libraries with mutations near the substrate binding pocket give often impressive results. Besides elaborated methods for preparing such libraries, efficient screening procedures are also crucial for project success. An example on library design and screening using new technologies will be given based on the engineering of an aspartase.
