Zusammenfassung
Massenspektrometrische Verfahren verfügen über ein großes Potenzial für die Bestimmung von Proteinen in Lebensmitteln. Die Kombination von Flüssigchromatografie und massenspektrometrischen Fragmentierungstechniken kann dazu genutzt werden, eine Vielzahl an Proteinen simultan in einem Lebensmittel zu bestimmen, sei es zur Authentizitätsprüfung oder zum Nachweis von Allergenen. Jedoch benötigt man für einen routinemäßigen und qualitätsgesicherten Einsatz in der Lebensmittelüberwachung standardisierte Methoden. Aus diesem Grund wurde die § 64 LFGB Arbeitsgruppe „Massenspektrometrische Proteinanalytik“ gegründet. Die Ziele der Arbeitsgruppe sind die Aufnahme von validierten Flüssigchromatografie-Massenspektrometrie-Methoden in die „Amtliche Sammlung von Verfahren zur Probenahme und zur Untersuchung“ und die Entwicklung von Leitfäden für die Validierung dieser Methoden. Am 13. März 2018 fand auf Einladung des Bundesamts für Verbraucherschutz und Lebensmittelsicherheit in Berlin das erste Treffen der Arbeitsgruppe statt. Auf der Sitzung wurden Methoden zur Proteinbestimmung in Lebensmitteln mittels LC-MS vorgestellt. Anschließend diskutierten die Mitglieder der Arbeitsgruppe über das Anwendungspotenzial der Methoden und über mögliche Validierungskonzepte. Für eine Methode zur Authentizitätsprüfung von Fischproben und für eine Methode zum Nachweis von Schalenfrüchten und Erdnuss wurde beschlossen, eine Überprüfung der Methoden durch laborübergreifende Vorringversuche durchzuführen, um daran anschließend Methodenvalidierungsstudien mit mindestens acht Laboren zu realisieren und auszuwerten.
Abstract
Mass spectrometric methods have a great potential to determine proteins in food. The combination of liquid chromatography and mass spectrometric fragmentation techniques can be used to determine a variety of proteins simultaneously in food, for both verification of authenticity or allergen detection. Method standardization is the key for the application of such technology in quality-assured routine analysis. For this reason, the § 64 LFGB working group “Mass Spectrometric Protein Analysis” was founded. The aim of the working group is to incorporate validated liquid chromatography mass spectrometry (LC–MS) methods to the “Official Collection of Methods of Analysis and Sampling” and to develop guidelines for the validation of such methods. The first meeting of the working group was held in March 2018 in Berlin on the invitation of the Federal Office of Consumer Protection and Food Safety. During the meeting, various methods for protein determination in food using LC–MS were presented. Subsequently, the members of the working group discussed the application potential of the methods and possible validation concepts. Two methods, one for identification of fish species, the other for detection of nuts, were proposed for an interlaboratory pre-ring trial evaluation, followed by the implementation and evaluation of method validation studies with at least eight laboratories.
