Immunogold detection of chromatophore antigens on the surface ofRhodopseudomonas sphaeroides spheroplasts

Abstract
Antibody was obtained from rabbits immunized with chromatophores isolated fromRhodopseudomonas sphaeroides. Antichromatophore antibody was adsorbed with membranes prepared from nonphotosynthetic aerobically grown cells to remove cross-reacting antibody in order to obtain an antibody preparation specific for antigens unique to the chromatophore membrane. This adsorbed antibody was coupled to colloidal gold particles and used to label chromatophore antigens on the surface of spheroplasts of phototrophically grown cells. Aerobically grown cells were induced to form chromatophore membrane by reduction of oxygen tension. Chromatophore-specific antigens were recognized by crossed immunoelectrophoretic analysis of membranes prepared from these induced cells and by immunogold labeling of spheroplasts prepared from these cells.
