Phylogenetic placement of the northeastern Pakistani Ajania fruticulosa (Ledeb.) Poljakov (Asteraceae) on the basis of nrDNA-ITS sequences

Abstract

This study was carried out to have a closer investigation on taxonomic position of the northeastern Pakistani Ajania fruticulosa with 3 genera viz., Artemisia, Ajania and Chrysanthemum. To achieve this, the nrDNA-ITS (nuclear ribosomal internal transcribed spacer) region was newly sequenced for A. fruticulosa (strain PMNH-041623) and pooled with already deposited ITS sequences in the GenBank from the three genera. In the resulting ITS phylogenetic maximum likelihood (ML) tree, A. fruticulosa specified close resemblance and appeared in a clade encompassing Ajania and Chrysanthemum species. The ITS phylogeny with an emphasis on three genera using ML approach proposed that Ajania and Chrysanthemum have close affinities with each other that might create confusions in their generic circumscription. Nevertheless, the monopyly of Artemisia genus was evident in the current ITS phylogeny. On the basis of outcomes of this study, it is proposed that bigger revised phylogenies with large number of taxa with the inclusion of northeastern Pakistani A. fruticulosa are necessary to clarify species delimitation and the intergeneric relationships in Ajania, Artemisia and Chrysanthemum. It is further recommended that comprehensive field surveys should be accompanied to acknowledge the distribution of more Ajania species across the northeastern Gilgit-Baltistan Pakistan.
