Abstract
Covalent DNA-protein crosslinks (DPCs) are generally toxic DNA lesions that must be repaired to prevent premature aging and cancer formation. However, DPCs formed by the protein HMCES are important to stabilize abasic sites during replication, thereby preventing the formation of dangerous DNA breaks. Here, we discuss the principles of HMCES-DPC formation and resolution, which encompasses both proteolytic and non-proteolytic mechanisms.
